چکیده

BACKGROUND Giardiasis is an intestinal infection correlated with poverty and poor drinking water quality, and treatment options are limited. According to the Center for Disease Control and Prevention, Giardia infections afflict nearly 33% of people in developing countries, and 2% of the adult population in the developed world. This study describes the single cyclic nucleotide-specific phosphodiesterase (PDE) of G. lamblia and assesses PDE inhibitors as a new generation of anti-giardial drugs. METHODS An extensive search of the Giardia genome database identified a single gene coding for a class I PDE, GlPDE. The predicted protein sequence was analyzed in-silico to characterize its domain structure and catalytic domain. Enzymatic activity of GlPDE was established by complementation of a PDE-deficient Saccharomyces cerevisiae strain, and enzyme kinetics were characterized in soluble yeast lysates. The potency of known PDE inhibitors was tested against the activity of recombinant GlPDE expressed in yeast and against proliferating Giardia trophozoites. Finally, the localization of epitope-tagged and ectopically expressed GlPDE in Giardia cells was investigated. RESULTS Giardia encodes a class I PDE. Catalytically important residues are fully conserved between GlPDE and human PDEs, but sequence differences between their catalytic domains suggest that designing Giardia-specific inhibitors is feasible. Recombinant GlPDE hydrolyzes cAMP with a Km of 408 μM, and cGMP is not accepted as a substrate. A number of drugs exhibit a high degree of correlation between their potency against the recombinant enzyme and their inhibition of trophozoite proliferation in culture. Epitope-tagged GlPDE localizes as dots in a pattern reminiscent of mitosomes and to the perinuclear region in Giardia. CONCLUSIONS Our data strongly suggest that inhibition of G. lamblia PDE activity leads to a profound inhibition of parasite proliferation and that GlPDE is a promising target for developing novel anti-giardial drugs.

Crystal Structures of Carbamate Kinase from Giardia lamblia Bound with Citric Acid and AMP-PNP

The parasite Giardia lamblia utilizes the L-arginine dihydrolase pathway to generate ATP from L-arginine. Carbamate kinase (CK) catalyzes the last step in this pathway, converting ADP and carbamoyl phosphate to ATP and ammonium carbamate. Because the L-arginine pathway is essential for G. lamblia survival and absent in high eukaryotes including humans, the enzyme is a potential target for drug ...

ZINC DEFICIENCY IN CHILDREN INFECTED WITH GIARDIA LAMBLIA

Giardia, a protozoan parasite, resides in the small intestine in man. This parasite damages the mucosal intestinal surface and in severe cases produces malabsorption and it may also affect mineral absorption, especially zinc. In this study a total of 118 infested and 43 non-infested persons as a control group were selected. From each patient a 5 ml blood sample was obtained. The serum was ...

Detection of Giardia lamblia Cysts in Surface Waters of Rasht City, Iran

Introduction: Giardia lamblia is a protozoan parasite with universal distribution in human populations. This infections transfer to human via contaminated foods and waters with Giardia cysts. The Knowledge on the incidence of this agent in the potential infection sources can provide valuable information for control and the spread of this parasite to human communities. This study was aimed to is...

Interleukin-6-deficient mice are highly susceptible to Giardia lamblia infection but exhibit normal intestinal immunoglobulin A responses against the parasite.

In the present study, interleukin-6 (IL-6)-deficient mice were infected with Giardia lamblia clone GS/M-83-H7. Murine IL-6 deficiency did not affect the synthesis of parasite-specific intestinal immunoglobulin A. However, in contrast to wild-type mice, IL-6-deficient animals were not able to control the acute phase of parasite infection. Reverse transcription-PCR-based quantitation of cytokine ...

Diagnostic Potential of Target Giardia lamblia Specific Antigen for Detection of Human Giardiasis Using Coproantigen Sandwich ELISA

Giardiasis is endemic in all regions of the world. Giardia lamblia (G. lamblia) cysts are spread in Egypt via the fecal-oral route, through ingestion of the cyst with contaminated food or water. The symptoms of giardiasis vary from the asymptomatic passage of cysts to chronic diarrhea, malabsorption and weight loss. Although microscopy has the advantage of low cost and ability to simultaneously...
